Even if the operator has strict technical specifications in the cell culture process, but there is no suitable culture environment, the risk of contamination will increase when the environment deteriorates. Therefore, the cell culture environment must be clean. One of the main sources of cell contamination is the constant temperature and humidity incubator.

In the constant temperature and humidity incubator, the cultured cells are in direct contact with the air during the removal and transfer process, entraining microorganisms; in addition, the internal humidity is high and the temperature is suitable, which is especially conducive to the reproduction of fungi. If the cell culture flask or Petri Dish comes into contact with anything other than the operating table, it must be wiped with sterile alcohol before placing it in the incubator.However, it should be noted that the disinfectant alcohol must be dried on the operating table, otherwise the ethanol concentration in the incubator will increase, and the alcohol will affect the normal function of the cells.

In order to effectively control this exposure route, the following measures can be taken:

1) Add antifungal agents such as copper sulfate and sodium dodecylsulfonate to the humidification tray to inhibit the growth of fungi in the tray; or keep sterile deionized water, change it once a week, and clean it with strict alcohol or high temperature Sterilize the trays;

2) Regularly clean the inside and outside of the incubator with a non-toxic and anti-fungal cleaner. Pay special attention to non-detachable corners;

3) Improper use of the air filter will become a terrible pollution path for the incubator, so the air filter needs to be replaced regularly according to the instructions;

4) During use, if there is any spillage, it must be cleaned up immediately with disinfectant alcohol;

5) Once the culture is found to be contaminated, clean it up immediately.
